В работе использованы клетки глиомы крысы линии С6 и клетки аденокарциномы шейки матки линии HeLa. Стимуляцию проводили на протяжении 12 ч однородным электрическим полем с частотой 10 Гц. Для измерения величины равновесного трансмембранного потенциала применяли метод пэтч-кламп, пролиферативную активность оценивали по изменению числа клеток относительно их числа в контрольном образце. Показано, что электрическая стимуляция вызывает изменение равновесного трансмембранного потенциала клеток, что приводит к последующему изменению их пролиферативной активности. Выявлено, что деполяризация плазматической мембраны усиливает пролиферативную активность клеток, в то время как гиперполяризация замедляет. Установлено, что конечный результат зависит как от параметров электрического поля, так и от типа клеток. Таким образом, наблюдаемый эффект связан с активацией потенциал-зависимых ионных каналов, приводящей к изменению времени прохождения клеткой определенной фазы клеточного цикла. Полученные результаты могут быть использованы для разработки методов клеточной инженерии. </p></abstract><trans-abstract xml:lang="en"><p>The influence of long-term electrical stimulation on the functional properties of cells in culture was investigated. C6 rat glioma cells and HeLa human adenocarcinoma cells were used in this work. Stimulation was carried out with uniform electric field for 12 hours with the train of pulses at a frequency of 10 Hz. Measurements of membrane potential were conducted using patch-clamp technique; the proliferative activity was evaluated by cell counting relative to control sample. It was shown that electrical stimulation causes a change in membrane potential of the cells and subsequent changes in their proliferative activity. It was established that membrane depolarisation increases proliferative activity whereas hyperpolarization decreases it. It was established that the effect depends on the parameters of the electric field and the type of cells. The observed effect is associated with activation of potential-dependent ion channels, resulting in changes of the specific phase duration in the cell cycle.
